1. Drugs that will increase DA activity and drugs that will decrease cholinergic activity
2. Amantadine acts as a metaffinoid potentiator of DA
3. (i) Pramipexole is an affinity for D3 receptors and decrease affective symptoms
(ii) Ropinirole is a D2 agonist and helps with severe heart failures
(iii) Bromocriptine acts together with L-dopa or anticholinergic drugs, for patients who cannot tolerate L-dopa.
(iv) Rotigotine also increases the DA level
4. Ergot - Bromocriptine, Not ergot - Pramipexole & ropinirole
5. Pramipexole and ropinirole, these two drugs are currently on the first line with initial treatment of Parkinson's disease in younger patients.
6. Monoamine Oxidase B inhibitor (they decrease the disease progression) & Pramipexole (they decrease affective symptoms)
7. MAO-B increases DA concentration in CNS, selegiline is an irreversible inhibitor of MAO-B & Safinamide is a potent reversible inhibitor of MAO-B.
8. COMT-inhibitor increase plasma levels of 3-O-methyl dopa weak therapeutic response with L-dopa. And it extend the duration of action of L-dopa, decrease peripheral metabolism & improve bioavailability.
9. Istradephylline is an adenosine A2 antagonist & add-on therapy to L-dopa/carbidopa experiencing off-episodes.
10. Safinamide:
Increase DA activity (potent reversible inhibition of MAO-B & inhibition of DA uptake)
Decrease glutamide release.
